Try to deactivate all third party plugins and check if this solves the issue - if yes activate them one by one to find the conflicting one. When I select the 3 menus from the pop downs on the 'Theme Location' option and save it seems to save but if I go to the front end it is still using the fallback menu structure. The main issue you will run into using the layout you have in your example is that your images are already 216x216 which means that on the mobile views they will need to stretch to expand and fill the container.
