May I please ask you if I have to add a thumbnail to a featured video that's set to "Big image/video" in the slideshow, in order to get a thumbnail to show up in the related posts?

Yes. When you link a video, the theme will initially display an icon of reels of film with the words 'click to define preview image.' Without that image, the related posts will show the same icon of movie reels but without the instructions.

When it comes to creating a thumbnail for a video that is full width (Big Image/Video slideshow) should the size of the preview image be the same size as video being played in the slide show?

It should have the same aspect ratio but if you make it too small it will wind up looking blurry. This is something you need to play around with to see what works best for you, because if you preview image is monotone with a vector icon as compared to a painting of a sunset, you would need to try a bunch of different sizes to get everything optimized. I wouldn't go too crazy with detail or mobile users may experience difficulties loading 20 different gigantic preview images.
Test 3 preview images made in identical proportion to the video at different sizes . Here is a url to a calculator to help you out, ( http://andrew.hedges.name/experiments/aspect_ratio/ ) just plug in the dimensions of the video and then plugin either the height or the width (not both, but has to be one) and it will spit out the second dimension that you would use to make an image with the same aspect ration as the original.
